I'd put a pack together that weighs 30-35 pounds and plan a route that involves 20 miles per day and see how it effects your body. Blisters will certainly be an issue and if you know where you are susceptible to them before you're under way on the big hike, you can address how to deal with them. I ended up with arch supports which made a huge difference. Also, if you're here in the west, you should hike at altitude and see how it effects you. ..
